α2-Macroglobulina, inhibidor de proteinasas altamente sofisticado

Resumen


A la superfamilia de las α-macroglobulinas pertenece la α2-Macroglobulina y la PZP, las cuales conforman el subgrupo de las α-macroglobulinas. Los componentes del complemento C-3, C-4 y C-5 hacen parte de esta superfamilia y conforman el subgrupo del complemento. Esta superfamilia tiene en común varios dominios como: el tiolester y el Terminal-C, con la excepción del C-5, el cual carece de este dominio. Todos estos dominios juegan un papel fisiológico muy importante, el tiolester como inhibidor de las proteinasas, uniéndolas de forma covalente al tiolester, la región cebadora que es hidrolizada por las proteínas, induciendo cambios conformacionales en la molécula y finalmente el Terminal-C que contribuye con la eliminación de los complejos de la circulación, uniéndose al receptor LRP/ α2-Macroglobulinas en la superficie de las células reticuloendoteliales. Este trabajo está basado en la α2- Macroglobulinas como una de las proteínas de esta superfamilia mas estudiada y además se le ha encontrado involucrada en muchas patologías como Alzheimer, Esclerosis múltiple, problemas de coagulación y fibrinólisis.
